Polo-like Kinase




Plk1 is a 601 amino acid protein that comprises an N-terminal kinase domain and a C-terminal regulatory domain that contains a sequence of WVSKWVDYSDKYGLGYQLCDNSVGVLFNDS termed the Polo box. The motif is only observed in the Polo-like kinases. Its function remains unknown. It may be involved in an auto-regulatory mechanism or in targeting the kinase to substrates. Deletion of the C-terminal regulatory domain results in activation of the kinase. Plks have a threonine in the activation segment whose phosphorylation is essential for activity.
