The main character, who was turned into a vampire by Cain. As a result, most of his mortal memories have been erased. He turned his small sister/step-daughter to be (see "Trivia") into a vampire, but she didn't want to live forever. She committed suicide by throwing herself through a window, burning with the sunlight. That is possibly the turning point in his life, when he decided to stop taking blood from unwilling humans. He is very strong, and can create a blood sword by biting his finger and bullets from his own blood drops. He is the owner of a detective office in the Kanagawa district of the city of Yokohama.
A member of an agency known as the N.O.S., she often retains Shido's help on nightbreed cases. Originally, she was horribly disfigured in a fire when she was eight, while her twin sister Katsumi was unharmed. It turned out that Kasumi was possessed by a nightbreed that wanted the girl whole. Kasumi was killed in a fire while trying to exorcise the night breed, but not before giving her soul... and her face... to Yayoi.
Yayoi is immune to the vampire's bite due to the presence of her older twin sister Kasumi's soul, so that she won't die or turn into a vampire when bitten by one. She usually feeds Shido with her own blood when he is hungry or weakened.
An orphaned girl whose parents were killed by nightbreeds. Shido "adopted" her. Cain had cut her severely and left her to bleed to death, and she asked Shido to be turned into a vampire because she didn't want to die, but to live with him forever. She turned into a vampire, but is not nearly as strong as Shido, and now she is struggling to reconcile her own humanity and her new powers. Yayoi feeds her as well.
Shido's assistant, a small, green "urban fairy." She often irritates the others with biting remarks. However, she also offers worthy advice and is often concerned whenever the team goes out on cases.
A power vampire who is much older than Shido. He turned Shido into a vampire many centuries before the anime's current events. He loves Shido and he is capable of doing anything to make Shido love him again.
Formless creatures that curse humans and are fed by their souls. They are low-class demons that live in shadow.
There are some other minor characters that appear in a single episode only.

- In the anime, vampires can go out during sunrise and sunset, but must protect their body with a hat or umbrella and dark clothes.
- Even if it is a horror anime, it has some soft Yaoi scenes with Shido and Cain.
- The original title of this series is Mayonaka no Tantei.
- The Golden Dawn, which is mentioned in the first "season" of the show (Episodes 1-5), is a forgotten plot point in the second season. It is speculated that the Golden Dawn was supposed to be children who were half-breed and half-human, as Cain's remarks hinted at.
- Some of the characters got a major remodeling between the first and second "season." For instance, in episodes 1-5, Cain was a dark-haired, red-eyed vampire. However, in episodes 7-12, Cain is drawn with long blond hair and golden eyes. Other characters were changed too, but not as dramatically as Cain.
- Unlike many anime series, Nightwalker did not originate from Manga , but from a Hentai video game by the same name, Mayonaka no Tantei. The anime itself is not hentai though.
- There has been some disagreement in the fandom as to whether the little girl in Shido's flashback was his little sister or his soon to be step-daughter. There is evidence pointing to the latter, though. In Japanese, Shido refers to the woman in the flashback as the girl's mother, not as their mother.
- Shido's memories changed a lot from one episode to the next. It's first shown that Cain forcibly turned Shido into a vampire. However, in a second set of flashbacks, it appears that Shido willingly let Cain bite him and turn him into a vampire.
